ABSTRACT

Objective:To evaluate the clinical outcomes of Chaput tuberosity osteotomy and lateral malleolus sagittal osteotomy in the treatment of talus osteochondral lesions with autologous osteochondral transplantation (AOT).Methods:From January 2017 to December 2019, Chaput tuberosity osteotomy and lateral malleolus sagittal osteotomy were performed in the AOT treatment of 11 patients with talus osteochondral lesions. They were 9 men 2 women, with a mean age of 32. 6 years (range, from 22 to 45 years). Their lesions were all Hepple type V. Their American Orthopaedic Foot and Ankle Society (AOFAS) score, visual analogue scale (VAS), International Knee Documentation Committee Knee Evaluation Form (IKDC) score, and imaging data were measured pre- and post-operatively for assessment of clinical outcomes.Results:All the patients were followed up for 13 to 24 months (average, 15.6 months). Their AOFAS (94.3±2.9) and VAS (1.2±0.4) scores at the final follow-up were significantly improved compared with the pre-operative values (49.5±6.6 and 5.7±1.2) ( P<0.05). There was no significant difference between their pre- and post-operative IKDC scores for the ipsilateral knee joint ( P>0.05). The imaging showed that the talus cystic change disappeared and the grafted bone was fully fused with the talus with no abnormal change in the joint space. Conclusion:In the AOT treatment of talus osteochondral lesions, Chaput tuberosity osteotomy and lateral malleolus sagittal osteotomy can obviously relieve ankle pain and improve ankle function.

ABSTRACT

This paper intends to systematically review the efficacy and imaging related studies of dialectical behavior therapy (DBT) in the treatment of non-suicidal self-injury (NSSI), so as to provide references for DBT treatment of NSSI. NSSI has a very high incidence in the population and is a major threat to the physical and mental health of adolescents. DBT shows certain advantages in the treatment of NSSI. Previous studies have preliminarily revealed the neurological effects of DBT treatment on patients, which provides an important objective basis for the efficacy of DBT treatment. In this paper, the efficacy of DBT in the treatment of NSSI and the brain imaging evidence are described and discussed, so as to provide references for the effectiveness of DBT in the treatment of NSSI.

ABSTRACT

Objective To investigate the influence of curcumin and its analogue H8 on glucose and lipid metabolism disorder in db/db mice. Methods The type 2 diabetes mouse model (db/db mice) was intragastrically administrated with curcumin and analogue H8 for 8 weeks.The blood biochemical indexes were measured.The expression of PEPCK and G6Pase mRNA was detected by real-time PCR in liver tissues.The expression of PEPCK and G6Pase protein was detected by Western blotting. Results Curcumin analogue H8 reduced blood glucose and lipids in db/db mice (P<0.01) and improved liver function related enzymes significantly.The levels of PEPCK and G6Pase mRNA in db/db mice were significantly decreased (P<0.01) and the expression levels of PEPCK and G6Pase protein were significantly decreased(P<0.01). Conclusion Curcumin analogue H8 improves the glucose and lipid metabolism disorder in db/db mice,and it is related to inhibiting the expression of PEPCK and G6Pase gene and protein.

ABSTRACT

Objective To study the impact of Xingnaojing (XNJ) injection (a preparation of Chinese herb medicine) on the permeability of blood-brain barrier and Caveolin-1 in cortex of brain after global ischemia-reperfusion.Methods Modified Pulsinelli method for four-vessel occlusion was employed to establish the global ischemia reperfusion model in rats.Male Wistar rats were randomly (random number) divided into three groups,namely sham group,model group and XNJ group.Each group was observed at 24 h,48 h and 72 h after ischemia reperfusion.The water content of brain tissue was determined by dry/wet weight ratio,while the Evans blue (EB) concentration in brain tissue was detected by spectrophotometer.Western blot was used to detect caveolin-1 level in the cerebral cortex.Results The water contents of brain tissue in model group and XNJ group were significantly higher than that in sham group 24 hour after ischemia-reperfusion (P < 0.05).But at 48 h and 72 h after ischemia-reperfusion,the brain water content in model group was significantly higher than that in XNJ group and sham group (P < 0.05).The EB concentrations in brain tissue in model group and XNJ group were higher than that in sham group 24 h after ischemia-reperfusion (P < 0.05).EB levels in sham group and XNJ group were significantly lower than that in model group 48 h and 72 h after ischemia-reperfusion (P < 0.05).Caveolin-1 levels in cerebral cortex in sham group and XNJ group were significantly higher than that in model group 24 h,48 h and 72 h after ischemia-reperfusion (P < 0.05).Conclusions After global ischemia-reperfusion,Xingnaojing (XNJ) injection could protect blood-brain barrier in virtue of regulating caveolin-1 protein level.

ABSTRACT

Male Wistar rats were randomly divided into sham group, model group, solvent group and XNJ group.The observations were conducted at the time points of 24 h, 48 h and 72 h after ischemia reperfusion.The water content of the brain tissues was determined by dry-wet weight method, while the Evans blue ( EB) content of brain tissue was detected by spectropho-tometry.The protein levels of ZO-1 in the cerebral cortex were analyzed by Western blot.RESULTS:The water contents in the brain tissues in model group, solvent group and XNJ group were significantly higher than those in sham group ( P<0.05) 24 h after ischemia reperfusion.However, the brain water contents in model group and solvent group were signifi-cantly higher than those in XNJ group and sham group (P<0.05) 48 h and 72 h after ischemia reperfusion.The EB con-tents in the brain tissues in model group, solvent group and XNJ group were entirely higher than that in sham group 24 h af-ter ischemia reperfusion (P<0.05).The EB contents in sham group and XNJ group were significantly lower than those in model group and solvent group 48 h and 72 h after ischemia reperfusion (P<0.05).The protein expression of ZO-1 in the rat cerebral cortex in model group, solvent group and XNJ group was significantly lower than that in sham group 24 h after ischemia-reperfusion (P<0.05).Similarly, 48 h and 72 h after ischemia reperfusion, ZO-1 protein level in the cortex in sham group and XNJ group was significantly higher than that in model group and solvent group (P<0.05).CONCLU-SION:At 48 h and 72 h after global ischemia-reperfusion, Xingnaojing injection play a protective role in blood-brain barri-er and this role may be associated with the increase in ZO-1 protein expression by Xingnaojing injection.

ABSTRACT

BACKGROUND:Mesenchymal stem celltransplantation promoted skin repair in trauma via various regulatory mechanisms and inhibited scar formation. At present, many scholars believed that bioactive factors secreted by mesenchymal stem cells played an important role. OBJECTIVE:To investigate the effects of bone marrow mesenchymal stem cellconditioned medium on the proliferation and col agen synthesis of hypertrophic scar fibroblasts. METHODS:Human bone marrow mesenchymal stem cells and hypertrophic scar fibroblasts were isolated and cultured, and bone marrow mesenchymal stem cellconditioned medium was prepared. Hypertrophic scar fibroblasts were cultured in vitro with 12, 24, and 48 hour-col ected conditioned medium for 24 hours, which was compared with blank control group. The proliferation of cells was determined by CCK-8. Type I and type III col agen expression in hypertrophic scar fibroblasts was detected using real-time PCR. RESULTS AND CONCLUSION:Compared with the blank control group, 24 and 48 hour-col ected conditioned medium significantly inhibited the proliferation of hypertrophic scar fibroblasts (P<0.01), and also suppressed col agen synthesis of hypertrophic scar fibroblasts (P<0.01). Results suggested that bone marrow mesenchymal stem cellconditioned medium inhibited the proliferation and col agen synthesis of hypertrophic scar fibroblasts by secreting anti-fibrotic bioactive factors, which may provide new theoretical supports for celltherapy to reduce cutaneous scarring.

ABSTRACT

Objective To explore the independent prognostic value of plasma myeloperoxidase (MPO) in cardiovascular event of patient with stable angina pectoris.Methods Two hundred and forty-seven patients with stable angina pectoris (defined as 50% stenosis in any coronary vessel) were selected as our subjects and they all were followed up for 2 years.Plasma MPO and hsCRP concentrations were measured.The major adverse cardiovascular events (MACEs),including death,myocardial infarction and stroke,occurred in patients were recorded.Results All patients were divided into high MPO group (84 cases and plasma MPO concentrations over 320 mg/L) and low MPO group (163 cases,plasma MPO concentrations less than 320 mg/L).Compared with patients in low MPO group,the risk of occurring MACEs in high MPO group was higher(HR =1.67,95% CI =1.32-2.57,P < 0.05).There was positive correlation between plasma MPO and hsCRP concentrations (r =0.132,P < 0.05).Conclusion MPO level may serve as a better prediction index for adverse cardiovascular events of patients with Stable angina pectorisindependently.MPO has relationship with hsCRP concentrations.

ABSTRACT

This paper summarized the assessment model reform practiced in the postgraduate cell biology course for several years and successfully obtain a new testing method based on research papers re-porting and questioning. Before the assessment, each student searched and selected one required research paper to read and make a PowerPoint reporting document. During assessment , some students were ex-tracted to speak and comprehensive score, which was regarded as student's final examination results was made by teaching group. The rest of the students listened to speeches , questioned and filled the questions on the record sheet.Teaching group reported a score , which was regarded as part of student's final exami-nation result based on questioning record sheets and printed PowerPoint documents.This method can well evaluate students' scientific thinking and strengthen the training of scientific thinking of postgraduates.

ABSTRACT

Objective To compare the efficacy of sedation with dexmedetomidine versus midazolam in mechanically ventilated patients.Methods Sixty patients aged 20-64 yr,with body mass index 21-25 kg/m2,APECHE Ⅱ score 10-25,requiring 24 h of nechanical ventilation in intensive care unit (ICU),were randomly divided into 2 groups (n=30 each): midazolam group (group M) and dexmedetomidine group (group D).A loading dose of midazolam 0.05 mg/kg was injected intravenously,followed by infusion at 0.03-0.20 mg· kg-1 · h-1 in group M.A loading dose of dexmedetomidine 1 μg/kg was injected intravenously,followed by infusion at 0.2-0.7 μg· kg-1 · h-1 in group D.Ramsay sedation score was maintained at 2-4.The satisfaction of the doctors in ICU with the sedative efficacy,hypotension and bradycardia were recorded.Delirium was recorded starting from the begging of sedation to 2 h after the end of sedation.The emergence time and occurrence of falling asleep again within 2 h after waking were also recorded.Results Compared with M group,the satisfactory level of the doctors in ICU with the sedative efficacy was significantly increased,the emergence time was significantly shortened and the incidences of falling asleep again within 2 h after waking and delirium were significantly decreased (P < 0.05 or 0.01),and no significant change was found in the incidence of hypotension and bradycardia in group D (P > 0.05).Conclusion The efficacy of sedation with dexmedetomidine is better than that of midazolam in mechanically ventilated patients.

ABSTRACT

Recently, more research about the plant bioreactor expressing genes encoding human proteins was reported. In the present study, the cDNA of the human gene keratinocyte growth factor 2 (KGF2) was replaced with plant preferred codons by PCR, and the modified full-length cDNA was cloned into the plant expression vector pCAMBIA-YO containing the oil-body promoter. The fusion construct pCAMBIA-YO-KGF2 was transformed into Brassica napus by Agrobacterium tumefacien-mediated cotyledon transformation method. The transgenic seedlings were identified by PCR, Southern and western blot analysis all showed that KGF2 gene was successfully expressed in in transgenic Brassica napus.

ABSTRACT

OBJECTIVE To investigate the clinical distribution and resistance of Alcaligenes xylosoxidans,discussing risk factors of nosocomial infection and measures of prevention and treatment.METHODS We retrospectively analyzed 1323 isolated strains of Al.xylosoxidans in our hospital from Sep 2004 to Aug 2006.RESULTS For specimen from which Al.xylosoxidans was examined,sputum accounted for 99.85%,being the highest percentage.Al.xylosoxidans was especially distributed at respiratory ward(46.94%) and intensive care unit(ICU) ward(12.77%).Then was cardiovascular ward,endocrinology ward and hematology ward.The antibiotic drugs which had the high drug-resistance rate(i.e.more than 98%) were aminoglycoside(amikacin and gentamicin),quinolone(ciprofloxacin) and cefepime.But the drug-resistance rate to carbapenems,?-lactam/?-lactamase inhibitor combinations and the third generation cepholosporins(cefoperazone and ceftazidime) was less than 5%.CONCLUSIONS It is important for clinic to strengthen the disinfection for hospital environment and to use antibiotic drugs reasonably in order to control colonization and spread of Al.xylosoxidans in hospital.

Objective To study the effect of an immuno-targeted therapy drug Herceptin on apoptosis and cell cycle of overexpressing HER2 breast cancer cells.Methods Breast cancer cell line SKBR-3 was treated with Herceptin at most effective dosage and exposure time were selected by MTT assay.Then the apoptotic features,the apoptotic rate and the change of cell cycle of breast cancer cells were detected by fluorescent microscope(FM),laser confocal microscope(LCM),scanning electron microscope(SEM),transmission electron microscope(TEM)and flow cytometry(FCM).Results With the treatment of Herceptin,the visible apoptosis features of SKBR-3 cells were observed by FM,LCM,SEM and TEM.Compared to the control cell group,the rate of initial apoptotic cells detected by FCM with Annexin V/PI staining increased significantly(P

Objective To establish MSP (methylation-specific PCR) method for detecting bcl-2 gene, to study bcl-2 methylation in the development of breast cancer and the relationship between bcl-2 methylation and expression. Methods Designing the MSP primer of bcl-2 gene, the methylations in CpG island of bcl-2 gene 5'-regulatory region in 30 cases of normal, 25 cases of atypical hyperplasia(precancerous condition), 40 cases of lymphnode(-)(low-stage) and 45 cases of lymphnode(+)(high-stage) breast tissue were detected by using MSP. The expressions of bcl-2 were detected by using SP immunohistochemical technique. Results The positive rates of bcl-2 methylation in normal, atypical hyperplasia, lymphnode(-) and lymphnode(+) breast tissue were 10.0%,32.0%,40.0% and 53.3% respectively. Objective To investigate the relations between p53 and K-ras gene mutation in portal venous blood of gastric carcinoma patients and cancer metastasis. Methods p53 and K-ras gene mutation was detected with PCR-SSCP technology in 62 cases of gastric carcinoma. ABSTRACT

Objective To study the relationships between cell kinetics, apoptosis and expression, mutation of related genes in breast cancer. Methods Flow cytometry, immunohistochemical SP and PCR SSCP were used respectively to study DNA index(DI), S phase fraction(SPF), proliferation index(PI), apoptosis index(AI), expression of c erbB 2, Bcl 2, p53, PCNA, Ki67, TopoⅡ and mutation of p53 in 54 cases of breast cancer. Results High DI, aneuploidy rate, SPF, PI and AI were coincided with high expression of c erbB 2, p53, PCNA, Ki167 and TopoⅡ. Low DI, SPF, PI and AI were coincided with high expression of Bcl 2. High DI, SPF, PI and AI were coincided with high mutation of p53. High AI was related to a type of p53 mutation——loss of heterozygosity(LOH). Conclusion Abnormal cell kinetics and apoptosis were related with abnormal expression, mutation of related genes in breast cancer. [
